چکیده

EXPRESSION IN THE ARCUATE NUCLEUS AND PARAVENTRICULAR NUCLEUS OF THE HYPOTHALAMUS by KARI ELIZABETH BOLT (Under the direction of Silvia Giraudo) ABSTRACT This study investigated the levels of mRNA of POMC, AgRP and MC4R in the arcuate nucleus and paraventricular nucleus of the hypothalamus in thirty Sprague Dawley rats fed a low fat diet or a high fat diet over the course of 12 weeks. The HF group had higher levels of POMC (p=0.013) and AgRP (p=0.001) in the ARC after 7 weeks of feeding. The HF group did have higher levels of MC4R (p=0.038) in the PVN after 12 weeks of feeding. POMC and AgRP mRNA expression in the ARC seemed to increase and decrease in a similar pattern during high fat feeding, while the varying levels of MC4R mRNA expression in the PVN seems to be inversely related to AgRP and POMC expression in the ARC. This may indicate a link in regulation or a breakdown energy balance that supports a state positive energy balance.

NPY and MC4R signaling regulate thyroid hormone levels during fasting through both central and peripheral pathways.

Fasting-induced suppression of the hypothalamic-pituitary-thyroid (HPT) axis is an adaptive response to decrease energy expenditure during food deprivation. Previous studies demonstrate that leptin communicates nutritional status to the HPT axis through thyrotropin-releasing hormone (TRH) in the paraventricular nucleus (PVN) of the hypothalamus. Leptin targets TRH neurons either directly or ind...

Melanocortin 4 receptor distribution in the human hypothalamus.

OBJECTIVE The melanocortin 4 receptor (MC4R) is an essential regulator of energy homeostasis and metabolism, and MC4R mutations represent the most prevalent monogenetic cause of obesity in humans known to date. Hypothalamic MC4Rs in rodents are well characterized in neuroanatomical and functional terms, but their expression pattern in the human hypothalamus is unknown. DESIGN AND METHODS To d...

The central melanocortin system and the integration of short- and long-term regulators of energy homeostasis.

The importance of the central melanocortin system in the regulation of energy balance is highlighted by studies in transgenic animals and humans with defects in this system. Mice that are engineered to be deficient for the melanocortin-4 receptor (MC4R) or pro-opiomelanocortin (POMC) and those that overexpress agouti or agouti-related protein (AgRP) all have a characteristic obese phenotype typ...

Brain nuclear receptors and body weight regulation

Introduction Obesity is a serious global health problem due to its increasing prevalence and comorbidities. Over 30% of Americans are obese (1). Obese individuals are at an increased risk for developing type 2 diabetes, cardiovascular disease, and cancer. These observations highlight the urgent need to better understand the physiology of body weight control, which may facilitate the development...

Neurochemical phenotype of hypothalamic neurons showing Fos expression 23 h after intracranial AgRP.

Agouti-related protein (AgRP) is coexpressed with neuropeptide Y (NPY) in a population of neurons in the arcuate nucleus (ARC) of the hypothalamus and stimulates food intake for up to 7 days if injected intracerebroventricularly. The prolonged food intake stimulation does not seem to depend on continued competition at the melanocortin-4 receptor (MC4R), because the relatively specific MC4R agon...
